Ксенобиотики обладают способностью изменять уровень экспрессии генов, степень которой зависит от генотипа. К ним относится винилхлорид (ВХ). Его воздействие вызывает изменения показателей липидного обмена проатерогенного характера.</p></sec><sec><title>Материал и методы</title><p>Материал и методы. Проведено проспективное когортное обследование 120 лиц мужского пола, контактирующих с ВХ (средний возраст – 46,9 ± 0,9 года; средний стаж – 17,8 ± 0,9). Исследовали показатели липидного обмена, результаты сравнивали с референтными значениями. Оценивали генотипы гена аполипопротеина С3 (ApoC3) C3238G rs5128, аполипопротеина Е (ApoE) Leu28Pro rs769452 и липопротеиновой липазы (LPL) Ser447Ter rs328. Оценку значимости межгрупповых различий и соответствие частот встречаемости генотипов закону Харди–Вайнберга осуществляли по критерию χ2. Ассоциацию аллелей или генотипов с предрасположенностью к нарушениям оценивали по величине отношения шансов (ОR).</p></sec><sec><title>Результаты</title><p>Результаты. У рабочих, экспонированных ВХ, увеличение концентрации проатерогенной фракции холестерина ассоциировано с носительством генотипа G/G полиморфного варианта С3238G гена АРОС3, протективным эффектом в отношении данного нарушения обладает каждый вариантный аллель полиморфного варианта Ser447Ter гена LPL. Носительство любого из вариантных аллелей C/G или G/G полиморфного варианта С3238G гена АРОС3 увеличивает вероятность развития нарушений уровня антиатерогенной фракции холестерина, а любого из вариантных аллелей T/C или C/C полиморфного варианта Leu28Pro гена АроЕ – уменьшает его. Вероятность повышения уровня триглицеридов увеличивается в присутствии обоих аллелей полиморфного варианта С3238G гена АРОС3 и снижается в случае носительства обоих аллелей полиморфного варианта Leu28Pro гена АроЕ. Наличие любого вариантного аллеля данного полиморфного варианта является значимым у лиц с повышенным уровнем общего холестерина.</p></sec><sec><title>Заключение</title><p>Заключение. В условиях воздействия ВХ происходит увеличение вероятности развития нарушений липидного обмена, ассоциированной с генами rs5128 и rs769452. Протективным действием обладает ген rs328.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. Xenobiotics having the ability to change the gene expression level, the degree of which depends on the genotype include vinyl chloride (VС). Its impact causes changes in lipid metabolism proatherogenic IN character. </p></sec><sec><title>Material and methods</title><p>Material and methods. A prospective cohort study was conducted In 120 males contacting with VС (mean age - 46.9±0.9 years; mean experience - 17.8±0.9 years). Lipid metabolism indices were studied; the results were compared with reference values. There were evaluated the genotypes of apolipoprotein C3 (ApoC3) C3238G rs5128, apolipoprotein E (ApoE) Leu28Pro rs769452 and lipoprotein lipase (LPL) Ser447Ter rs328. The significance of intergroup differences and correspondence of genotype frequencies to the Hardy – Weinberg law were assessed by the criterion χ2. Association of alleles or genotypes with a predisposition to disorders was assessed by odds ratio (OR). </p></sec><sec><title>Results</title><p>Results. In workers exposed to VC, there was an increase of the concentration of proatherogenic cholesterol fraction associated with carriage of genotype G/G polymorphic variant of the gene С3238G АРОС3, protective effect against this violation has each variant allele of a polymorphic variant of the gene LPL Ser447Ter. Carriers of any variant allele C/G or G/G polymorphic variant of the gene С3238G АРОС3 increases the likelihood of violations the antiatherogenic cholesterol fraction level, and any of the variant alleles T/C or C/C polymorphic variant of the gene Leu28Pro ApoE – reduces it. Probability of an increase the triglycerides level is elevated in the presence of both alleles of polymorphic gene variant of a gene С3238G АРОС3 and decreases – in the case of carriers of both polymorphic alleles of the gene variant Leu28Pro АроЕ. The presence of any variant allele of this polymorphic variant is significant in individuals with elevated total cholesterol. </p></sec><sec><title>Conclusion</title><p>Conclusion. In terms of exposure to VC the probability of development of disorders of lipid metabolism-associated with genes rs5128 and rs769452.
