Микропластик представляет серьёзную угрозу здоровью из-за токсичности и способности к биоаккумуляции. Исследования подтверждают его негативное влияние на организм, однако механизмы накопления и распределения частиц изучены недостаточно. Данное исследование направлено на разработку и экспериментальную проверку полуколичественного метода оценки этих процессов.</p><p>Цель исследования – разработка и апробация в условиях острого эксперимента полуколичественного метода оценки распределения и накопления микрочастиц пластика в организме.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Для исследования использовали 12 самок крыс линии Wistar, разделённых на четыре группы. Животным внутрисердечно вводили суспензию микрочастиц полистирола 50 мкл (диаметром 100; 500 и 1000 нм) или 50 мкл физиологического раствора. Накопление микропластика в тканях трёх органов оценивали полуколичественным методом с использованием флуоресцентной микроскопии и ранговой шкалы. Данные анализировали методом Bootstrap с поправкой Холма – Бонферрони, значимость различий определялась при p &lt; 0,05.</p></sec><sec><title>Результаты</title><p>Результаты. Микрочастицы пластика размером 100; 500 и 1000 нм при внутрисердечном введении обнаруживаются в печени, почках и лёгких, локализуясь в печёночных триадах, корковом слое почек и областях ацинусов, альвеолярных ходах лёгких, с наибольшим количеством при введении частиц размером 1000 нм. Полуколичественная оценка выявила, что наибольшее накопление микрочастиц пластика в печени и лёгких наблюдается при введении частиц размером 1000 нм, в почках – при введении частиц 100 нм.</p></sec><sec><title>Ограничения исследования</title><p>Ограничения исследования. Исследование было ограничено изучением распределения микрочастиц пластика трёх размеров в токсикологическом эксперименте на одном виде животных (лабораторная крыса, по три животных в каждой группе).</p></sec><sec><title>Заключение</title><p>Заключение. Показано, что интенсивность и количество флуоресцирующих тканевых элементов зависит от размера частиц и могут быть связаны с образованием конгломератов из микрочастиц полистирола. Метод полуколичественной оценки позволил установить некоторые закономерности распределения изученных микрочастиц, но требуются дополнительные исследования с разработкой количественного подхода для повышения точности результата.</p><p>Соблюдение этических стандартов. Все эксперименты в рамках исследования соответствовали этическим нормам обращения с животными, принятым Европейской конвенцией по защите позвоночных животных, используемым для исследовательских и иных научных целей, Директивой Европейского парламента и Совета Европейского союза 2010/63/ЕС от 22.09.2010 г. о защите животных, использующихся для научных целей. Microplastics pose a significant health threat due to their toxicity and capacity for bioaccumulation. Although studies have confirmed their detrimental effects on biological systems, the mechanisms governing particle accumulation and particle distributions have not been sufficiently studied. </p><p>The aim of our study was to develop and experimentally validate a semi-quantitative method to assess these processes on the base of investigation of the distribution of plastic microparticles in the organism under acute in vivo experimental conditions using a semi-quantitative method.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Twelve female Wistar rats were divided into four groups. Animals received intracardiac injections of polystyrene microparticle suspensions (100, 500, and 1000 nm in diameter) or physiological saline solution. The accumulation of microparticles in six organs was evaluated using a semi-quantitative scoring method based on fluorescence microscopy and a rank-scale assessment. Data were analyzed using bootstrap methods with Holm–Bonferroni corrections, and differences were considered significant at p &lt; 0.05.</p></sec><sec><title>Results</title><p>Results. Following intracardiac administration, polystyrene microparticles measuring 100, 500, and 1000 nm were detected in the liver, kidneys, and lungs. They localized primarily in hepatic triads, the renal cortex, and the acinar regions and alveolar ducts of the lungs. The highest levels were observed upon administration of 1000-nm particles. Semi-quantitative analysis revealed liver and lung tissues to accumulate significantly more microparticles of the 1000-nm in size, while the kidneys showed greater accumulation of the 100-nm particles.</p></sec><sec><title>Limitations</title><p>Limitations. This study was limited to examining the distribution of three microparticle size groups in a single animal model (laboratory rats, only 3 animals in each group) under acute toxicological conditions.</p></sec><sec><title>Conclusion</title><p>Conclusion. The intensity and abundance of fluorescent tissue elements depend on particle size and may be associated with the formation of microparticle conglomerates. While the semi-quantitative method allowed identifying certain distribution patterns, further research involving quantitative approaches is required to enhance the accuracy and specificity of these findings.</p><p>Compliance with ethical standards.
