Недоочищенные сточные воды несут большую угрозу здоровью людей и окружающей среде. Не все очистные сооружения эффективно справляются со своей задачей.</p><p>Цель работы – сравнение патогенного потенциала бактериальных изолятов семейства Enterobacteriaceae, выделенных из сточных и поверхностных вод Москвы после очистных сооружений (ОС).</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Отбор образцов воды проводили на ОС на этапах очистки (вход, после цикла очистки, сбросовый канал) и из поверхностных вод (выше и ниже по течению от точки сброса с ОС). Исследования проводили на ОС Москвы: Зеленоградских очистных сооружениях (ЗОС), очистных сооружениях «Южное Бутово» (ЮБОС), Люберецких очистных сооружениях (ЛОС) и Курьяновских очистных сооружениях (КОС). Для детекции потенциально патогенных детерминант использовали метод ПЦР.</p></sec><sec><title>Результаты</title><p>Результаты. Установлено, что изоляты, выделенные из исследованных образцов воды ОС, имеют более высокий патогенный потенциал, чем изоляты, выделенные из кала. Гены устойчивости к антибиотикам встречаются у 7,5% изолятов E. coli, выделенных только из образцов воды ОС. Патогенные детерминанты достоверно чаще встречались в изолятах из образцов воды ОС (mrkD и kfu – для Klebsiella, aggR и ipaH – для E. coli). Все станции снижали количество изолятов после цикла очистки (для E. coli – в 2–11 раз, для клебсиелл – в 2–4 раза), кроме ЮБОС, где количество изолятов клебсиелл снижалось всего на 15%, а E. coli даже возрастало на 14%.</p></sec><sec><title>Ограничения исследования</title><p>Ограничения исследования. Изучали изоляты семейства Enterobacteriaceae из ОС Москвы и кала людей (К), из рода Klebsiella (157 СВ и 117 К-изолятов) и Escherichia coli (100 СВ и 80 К-изолятов) изолятов. Выборка ограничена по географии, поэтому выводы можно применять к ОС Москвы и Московской области (или сходных географических и экологических условий), где используют сходные схемы очистки.</p></sec><sec><title>Заключение</title><p>Заключение. В целом в сточных водах (в том числе неочищенных) из ОС Москвы выделены бактериальные изоляты семейства Enterobacteriaceae, имеющие больший патогенный потенциал, чем у изолятов, выделенных из кала людей. При этом установлено, что в результате очистки воды на очистных сооружениях Москвы (кроме ЮБОС) снижается на четыре порядка уровень бактериального загрязнения сточных вод представителями бактерий семейства Enterobacteriaceae, однако у жизнеспособных изолятов, выделенных в сбросном канале и в поверхностных водах ниже спуска сточных вод из ОС, снижение патогенного потенциала не зафиксировано.</p><p>Соблюдение этических стандартов. Исследование биологического материала от людей было одобрено Локальным независимым этическим комитетом (протокол № 98А заседания Локального независимого этического комитета ФГБУ «ГНЦК им. А.Н. Untreated wastewater poses a significant threat to public health and the environment. Not all treatment facilities are equally effective. </p></sec><sec><title>Purpose of the study</title><p>Purpose of the study. Comparison of the pathogenic potential of bacterial isolates of the Enterobacteriaceae family isolated from wastewater and surface water after treatment facilities (TF) in Moscow.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. Water samples were collected at the TF (inlet, post-treatment, and discharge) and from surface waters (upstream and downstream of the TF discharge point). Studies were conducted at Moscow’s TF: Zelenograd Wastewater Treatment Plant (ZWTP), Yuzhnoye Butovo Wastewater Treatment Plant (YUBOTWTP), Lyubertsy Wastewater Treatment Plant (LOS), and Kuryanovsk Wastewater Treatment Plant (KOS). PCR was used to detect potentially pathogenic determinants.</p></sec><sec><title>Results</title><p>Results. Isolates recovered from the analyzed water samples at the TF were established to have a higher pathogenic potential than isolates isolated from feces (F). Antibiotic resistance genes were found in 7.5% of E. coli isolates isolated only from the TF. Pathogenic determinants were significantly more common in isolates from the TF (mrkD and CFU for Klebsiella, aggR and ipaH for E. coli). All stations reduced the number of isolates after the treatment cycle (2–11 times for E. coli and 2–4 times for Klebsiella), except for YUBOS, where the number of Klebsiella isolates decreased by only 15%, while E. coli even increased by 14%.</p></sec><sec><title>Limitations</title><p>Limitations. We studied isolates of the Enterobacteriaceae family from Moscow TF and human feces (F) from the genus Klebsiella (157 wastewater and 117 F isolates) and Escherichia coli (100 wastewater and 80 F isolates), which represents a sufficient reference sample.</p></sec><sec><title>Conclusion</title><p>Conclusion. Overall, bacterial isolates of the Enterobacteriaceae family have been isolated from wastewater (including untreated wastewater) from Moscow’s wastewater treatment plants (WWTPs) and have a higher pathogenic potential than isolates recovered from human feces. Furthermore, water treatment at Moscow’s wastewater treatment plants (except for the YUBOTWTP) has been shown to reduce the level of bacterial contamination of wastewater by Enterobacteriaceae by four orders of magnitude. However, no reduction in pathogenic potential has been observed in viable isolates recovered from the discharge channel or from surface waters downstream of the WWTPs discharge.</p><p>Compliance with ethical standards.
