Хронический стресс и воздействие парацетамола относятся к наиболее актуальным факторам, влияющим на нейрохимические и молекулярные процессы в мозге современного человека. Избыточная активация воспалительных и окислительных путей при хроническом психосоциальном стрессе может приводить к снижению экспрессии нейропротекторных и антиоксидантных генов, усугубляя нейротоксические эффекты лекарственных средств, в том числе парацетамола.</p><p>Цель работы – оценить влияние хронического стресса и перорального введения парацетамола, а также их сочетанного воздействия на морфологическое состояние головного мозга, экспрессию генов антиоксидантной системы и нейротрофического фактора в гиппокампе крыс.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Эксперимент проведён на 48 крысах (линия Wistar, самцы и самки поровну), разделённых на четыре группы: контроль, хронический стресс, парацетамол, сочетанное воздействие стресса и парацетамола. Для моделирования хронического стресса использовали комплекс разнообразных стрессоров. Парацетамол вводили перорально в дозе 1000 мг/кг. Морфологическое состояние гиппокампа оценивали гистологически, экспрессию генов – методом количественной ПЦР в реальном времени. Статистический анализ включал бутстрэппинг и коррекцию множественных сравнений методом Холма – Бонферрони.</p></sec><sec><title>Результаты</title><p>Результаты. Макроскопическое и гистологическое исследование головного мозга крыс выявило сохранение нормальной анатомической структуры и целостности тканей гиппокампа во всех группах. Статистический анализ экспрессии генов показал, что у животных, получавших парацетамол (группы III и IV), наблюдается значительное снижение уровней мРНК генов Sod1 и Bdnf в гиппокампе по сравнению с контролем и группой, подвергшейся только стрессу (группа II). Уровень экспрессии Sod1 и Bdnf был существенно ниже как у самцов, так и у самок в этих группах (p = 0,001). Для гена Hmox1 значимое снижение экспрессии выявлено только у самцов в группе, получавшей парацетамол, тогда как у самок изменений не отмечено.</p></sec><sec><title>Ограничения исследования</title><p>Ограничения исследования. Для эксперимента были выбраны лабораторные животные одного биологического вида, токсикант использовали в одной концентрации.</p></sec><sec><title>Заключение</title><p>Заключение. Парацетамол и в изолированном применении, и в сочетании с хроническим стрессом оказывает более выраженное угнетающее действие на экспрессию ключевых генов антиоксидантной защиты и нейротрофической поддержки по сравнению с изолированным воздействием стресса.</p><p>Соблюдение этических стандартов. Дата заседания биоэтической комиссии ФБУН «Уфимский НИИ медицины труда и экологии человека» 08.02.2024 г. № 01–02. На протяжении всего эксперимента животные содержались в стандартных условиях: 12 ч искусственного освещения днём, влажность 30–70% и температура воздуха плюс 20–25 °C. Chronic stress and paracetamol exposure are among the most relevant factors affecting neurochemical and molecular processes in the brain in modern humans. Excessive activation of inflammatory and oxidative pathways during chronic psychosocial stress can lead to a decrease in the expression of neuroprotective and antioxidant genes, exacerbating the neurotoxic effects of drugs such as paracetamol.</p></sec><sec><title>Objective</title><p>Objective. To assess the effect of chronic unpredictable stress and oral administration of paracetamol, as well as their combined effect on the morphological state of the brain and on the expression of genes of the antioxidant system and neurotrophic factor in the hippocampus in rats.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. The experiment was conducted on forty eight rats (males and females in equal numbers), divided into four groups: control, chronic stress, paracetamol, combined exposure to stress and paracetamol. A complex of various stressors was used to model chronic stress. Paracetamol was administered orally at a dose of 1000 mg/kg. The morphological state of the hippocampus was assessed histologically, gene expression - by quantitative real-time PCR. Statistical analysis included bootstrapping and correction for multiple comparisons using the Holm-Bonferroni method.</p></sec><sec><title>Results</title><p>Results. Macroscopic and histological examination of the rat brain showed the preservation of the normal anatomical structure and integrity of the hippocampal tissues in all groups. Statistical analysis of gene expression showed animals treated with paracetamol (groups III and IV) to show a significant decrease in the mRNA levels in the Sod1 and Bdnf genes in the hippocampus compared to the control and the group exposed to stress only (group II). The expression level of Sod1 and Bdnf was significantly lower in both males and females in these groups (p = 0.001). For the Hmox1 gene, a significant decrease in expression was found only in males in the paracetamol-treated group, while no changes were noted in females.</p></sec><sec><title>Limitations</title><p>Limitations. Laboratory animals of the same biological species were used for the experiment, and the toxicant was used in only one concentration.</p></sec><sec><title>Conclusion</title><p>Conclusion. Thus, paracetamol, both in isolated use and in combination with chronic stress, has a more pronounced inhibitory effect on the expression of key genes of antioxidant protection and neurotrophic support than stress alone.</p><p>Compliance with ethical standards. Throughout the study, the animals were kept in standard conditions with 12-hour artificial lighting during the daytime, a relatively constant level of humidity (30–70%) and an air temperature of 20–25 °C.
